A Research Assistant position is available in the Process Development Lab (PDL), led by Marina Radrizzani at the San Raffaele Telethon Institute for Gene Therapy (SR-Tiget), Milan, Italy (https://research.hsr.it/en/institutes/san-raffaele-telethon-institute-for-gene-therapy/process-development-lab.html).The Institute. SR-Tiget was created in 1996 as a joint-venture between Fondazione Telethon and Ospedale San Raffaele (OSR), with the mission to perform cutting edge research on gene and cell therapy and to translate its results into therapeutic advances for genetic diseases. The Institute is located within the OSR campus, which includes a large multi-disciplinary research hospital, a biomedical research center, a university and also hosts several biotechnologies companies. Over the years, SR-Tiget has given a pioneering contribution to the gene and cell therapy field with relevant discoveries in vector design, gene transfer and gene editing strategies, stem cell biology, identity and mechanism of action of regulatory cells in immune responses and innate immune cells in cancer. SR-Tiget has also established the resources and framework for translating these advances into novel experimental therapies and has implemented several successful gene therapy clinical trials, which have already treated >130 patients and led to the filing and approval of 2 novel advanced therapy medicines. The position. We are looking for a motivated Research Assistant to work in the PDL Vector Core, which is focused on manufacturing of high-quality lentiviral vectors (LV) for in vitro and in vivo preclinical studies. Starting projects are aimed to improve our upstream and downstream process to develop a best-in-class scalable manufacturing platform, in order to increase LV yield and quality. In parallel, new analytical technologies will be explored.Main tasks-         Experimental activities in the context of-         Upstream LV manufacturing processes (flatware cultures and 3D bioreactors)-        Downstream LV manufacturing processes, based on chromatography, tangential flow filtration, direct filtration-         LV quality control: cell-based assays, molecular assays, flow cytometry analyses, ELISA-         Data analysis and reporting-         Preparation of standard lab procedures and protocolsQualifications and skills-         Master Degree  (Biology, Biotechnology or equivalent), PhD as a plus-         Strong wet lab experience-         Experience in Good Manufacturing Practices and/or LV production and quality control as a plus-         High accuracy, reliability, precision-         Friendly, collaborative, proactive mindset-         Good oral and written EnglishWe offer-         Competitive salary and contract depending on qualifications and experience-         A dynamic environment and the chance to give important contribution to innovative projects in the LV-based gene therapy fieldApplications, including a CV and the name and contact details of at least one referee, should be sent by e-mail to radrizzani.marina@hsr.it and montaldo.elisa@hsr.it.
